Abstract
Success with the baculovirus expression system is dependent on being able to infect cells efficiently with AcMNPV, thus obtaining maximum virus replication and hence optimum production of the desired protein. Before being able to create a recombinant virus in the first place you must have stocks of infectious, purified virus DNA into which you will insert the foreign gene. You will need to obtain the appropriate virus stock (AcMNPV or AcMNPY.lacZ) to amplify the virus and to purify virus particles from which the DNA can be isolated. All of these procedures are quite straightforward and relatively easy to undertake in the laboratory, if a few simple guidelines are adhered to.
