Salivary epithelium branching morphogenesis

Abstract

The embryonic rodent salivary gland has been used as a model for epithelial morphogenesis for over 40 years. During this period we have learned a great deal about the requirements for normal development in this organ. The submandibular salivary gland first appears late on day 12 of gestation in the mouse and on day 13 in the rat. The initially broad primordium arises from the oral epithelium and forms a stalk (primary duct) and a bulbous lobule. Over the next several days in vivo or in vitro the epithelium undergoes rapid growth and repetitive branching to generate a highly branched structure with greatly increased surface area (Figure 13.1). The submandibular gland has a highly characteristic branching pattern which is quite distinct from other branching organs such as the lung, mammary gland and pancreas (Figure 1; Spooner and Thompson-Pletscher, 1986).
