Abstract
An organism must be able to modulate the expression of its genes. Some proteins, such as the enzymes of key metabolic pathways and the ribosomal proteins, will be required in large quantities. In an E. coli cell it has been calculated that there are 20000 ribosomes, so it follows that there will need to be at least 20 000 copies of each of the constituent ribosomal proteins. Conversely, some proteins are normally present at very low concentrations but can be made in greater quantities when required. Thus, in E. coli growing on glucose, the enzyme β-galactosidase is normally present at approximately five molecules per cell but, under circumstances to be described in the following sections, it can be synthesized in a thousandfold greater quantities.
