Abstract

The rat pheochromocytoma (PC12) cell line is a widely used model system for nerve growth factor (NGF)-induced neuronal differentiation (1). Morphological differentiation is characterized by enlargement of cell bodies and extension of neurites. The mechanism of action of NGF is incompletely understood (2). However, in addition to morphological differentiation, several properties of PC12 cells change during exposure to NGF. Among others, densities of Ca2+ channels in the surface membrane are increased (3-6). This may have important functional consequences, for example for depolarization-evoked catecholamine release from the cells. It has been shown by several groups (7-9) that, in the course of differentiation, there is a change in sensitivity of catecholamine release to pharmacological interventions. The purpose of this paper is to summarize the evidence for this NGF-induced change.
