Abstract
To enhance recombinant protein productivities in animal cells by using oncogenes, we constructed reporter plasmids expressing the human interleukin-6 (hIL-6) gene under the regulation of various promoters, and effector plasmids containing various oncogenes fused with the cytomegallovirus (CMV) promoter. The results of transient expression experiments using COS-1 cells suggested that the CMV promoter is enhanced by the ras oncogene product. BHK-21 cells expressing the hIL-6 gene under the control of the CMV promoter were co-transfected with an effector plasmid and the dihydrofolate reductase gene then selected with 50 nM methotrexate to amplify the ras oncogene. We found that stable highly productive clones exhibiting about 20 times higher productivity rates compared to the control value could rapidly be established by employing this method.
