Abstract
Based on the method of full fractional factorial analysis we developed a fed batch assay to characterize the main energy metabolism of animal cells. The cells were cultivated in 6 well plates in different medium heights (2, 6 mm) with different glucose concentrations (5 g 1-1; 1 g 1-1) and glutamine concentrations (1 mmol 1-1; 4 nunol 1-1). Center point experiments (4 mm medium height, 2.75 g 1-1glucose, 2.5 mmol glutamine) allowed calculation of the experimental error and test for curvature of the observed parameter. The influence on the oxygen uptake rate (OUR), the growth rate and the maximum cell number of a mouse hybridoma (CD3) and the influence on the viability of human peripheral blood cells (PBC) is presented in this paper.
