Abstract
Rabbit polymorphonuclear leukocytes (PMNs) elicited by intraperitoneal injections of glycogen were prepared and suspended in Eagle’s MEM (Davidson et al. (1978). Br. J. Pharmacol. 63, 407P). PMNs were incubated in the presence and absence of serum opsonized zymosan (1mg/ml) and/or 10-5 mol/1 calcium ionophore (A23187) for 2 h at 37°C. Cells were removed by centrifugationa and aliquots of the supernatant removed for enzyme assays and radio-immunoassay of PGE2. The remaining supernatant was acidified and extracted with diethyl ether. The ether extracts were either used for bioassay of PGE-like activity (rat fundic strip) or derivatized to form the methyl ester, methoxime, tertiary butyldimethylsilyl ethers of the prostaglandins and TXB2 and the levels were measured by quantitative gas chromatography mass spectrometry (GC-MS).
