Abstract

The insight into the nature of the blue copper site in azurin that has been obtained using physical methods is briefly reviewed. The additional insight that has come from studies of the cobalt(II), nickel(II) and manganese(II) derivatives of the protein is then sketched. It is shown that in combination these results have yielded a resonable description of the electronic and geometric structure of the metal binding site. In accord with a recent crystallographic report, it is concluded that the site involves a cysteine sulfur, a methionine sulfur, and two imidazole nitrogens disposed in a pseudotetrahedral fashion.
