Abstract
The primary DNA adducts formed by metabolicallv activated aflatoxin B1 (AEB1), i.e. adducts released by acid hydrolysis as 2, 3-dihydro-2- (N7-guanyl)-3-hydroxyaflatoxin B1 (AFB1-N7 - and 37° and in situ in C3H 10T1/2 clone 8 mouse embryo fibroblasts. They undergo three major spontaneous reactions (1) release of 2,3-dihydro-2,3-dihydroxyaflatoxin B. resulting in the reconstitution of intact guanine residues; this reaction predominates under physiological conditions (2) release of free AFB.-N-Gua presumably under formation of aguaninic sites (3) Formation of secondary products in DNA which possess increased chemical stability probably by opening of the 5-membgred ring of guanine under formatiop of the putative 2,3-dihydro- 2-(N -formyl-2’,5’,6’-triamino-4’-oxo-H -pyrimidyl)-3-hydroxyaflatoxin B1 (AFB1-triamino-Py).
