SOP/DTX/150 Assay of DNA Repair Using [3H]-Deoxyguanosine
This assay is complementary to that described in SOP/DTX/149. Many chemicals preferentially damage purine residues within DNA particularly guanine residues. The repair of such damage can involve the replacement of only a few nucleotides at the site of the lesion. In many cases therefore, such repair may not involve the turnover of thymidine residues and hence will not be detected by the technique discussed in SOP/DTX/149. Thus, an assay using [3H]-deoxyguanosine is used when results by the first assay are negative or give doubtful positive responses.
