Abstract

The helical repeat of the DNA in the nucleosome is evaluated by three independent methods: from periodical distribution of the dinucleotides along known nucleosomal DNA sequences, from topology of DNA super-helix in the nucleosome and from analysis of accessibility of DNA in the nucleosome to DNase I, all giving similar results. All previous estimates are critically reviewed and the average figure is obtained, 10.38 ± 0.02 base-pairs per turn, based on five independent estimates. Experimental data available indicating that the free nucleosomal DNA is inherently curved, are reviewed. A novel approach to the higher order structure of chromatin is discussed which is based on steric exclusion effects imposing specific restrictions on the relative spatial positioning of the neighboring nucleosomes.
