Analysis of Individual Glucosinolates in Rapeseeds Comparison Between Different Methods

Abstract

The aim of our work is to contribute to the elaboration of a reliable quantitative method for individual glucosinolates determination in rapeseeds.

To emphasize the accuracy of the method, we have studied several important points:

  1. a)

    methanol - water (70%) extraction from seeds and seedcakes obtained after removal of the oil.

  2. b)

    column chromatographic purification of crude extracts

  • the influence of the height of D.E.A.E. A 25 columns (desulpho glucosinolates) and of Ecteolla Cellulose (intact glucosinolates) on the level of glucosinolates obtained is analysed

  • to avoid the use of pyridin, the pyridin - acetate buffer frequently used to prepare D.E.A.E. columns is changed by an acetic acid - sodium acetate buffer (pH : 5)

  1. c)

    HPLC analysis of desulphoglucosinolates and intact glucosinolates

  • comparison of desulphoglucosinolates and intact glucosinolates separations

  • choice of the detection wavelength

  • influence of column temperature

  1. d)

    Identification of desulphoglucosinolates from different cultivars of rapeseed (Garant, Ledos, Lirasol and Librador).
