Estrogen Receptor Immunocytochemical Assay (ER-ICA) and Laminin Detection in 115 Breast Carcinomas: A Light and Electron Microscopy Study with a Computerized (Samba 200) Quantitative Analysis on Tissue Sections

Abstract

Currently available histochemical or immunohistochemical methods (Dandliker et al., 1978; Kurzon and Sternberger, 1978; Lee, 1978; Nenci et al., 1976) have been shown to be unreliable for ER detection (Chamness et al., 1980). Recently, specific monoclonal antibodies against ER have been developed (Desombre et al., 1984; Greene et al., 1984; King and Greene, 1984; King et al., 1985; Press and Greene, 1984) making it possible to carry out either ER enzyme immuno-assays (ER-EIA) or ER immunocytochemical assays (ER-ICA) based on ER antigenic site detection. We undertook a prospective study in a large series of breast carcinomas using this new method with monoclonal anti-ER antibody (H 222S p)) in conjunction with an immunoperoxidase method. Our objective was to compare an ER immunocytochemical assay with current biochemical assays, to study variations in ER distribution in tumors using a computerized system for immunostaining analysis and, as accurately as possible, to delineate intracellular distribution of immunoreactive ER using immuno-electron microscopy.
