Abstract

Protoplasts derived from cell suspension cultures show a wide range of variations according to different parameters such as metabolic activity or DNA content (Lucretti et al., 1987; Carlberg et al., 1984). Flow-cytometry (FCM) has been applied for assessing the homogeneity of a potato cell suspension, concerning viability and trans-membrane potential as related to cell membrane integrity and functional mitochondria; for this purpose we used the cationic fluorescent dye rhodamine 123 (R123) specifically taken up by mitochondria of living cells (Johnson et al., 1981). The toxicity of the fluorochrome to the protoplast culture has been tested to establish the more convenient concentration to employ for FCM.
