Abstract

The application of tissue culture techniques to epithelial cells has revolutionized the study of morphologically complex or inaccessible tissues such as endocrine and exocrine glands, renal tubules, airway epithelia and ocular epithelial cells. Epithelial physiologists can now examine cell differentiation and other regulatory processes under precisely controlled conditions. When used in conjunction with molecular biological and other genetic methods, these fundamental processes can be assessed at the molecular level. This chapter focuses mainly on techniques for growing differentiated epithelial monolayers on permeable supports. This approach can yield morphologically simple monolayer epithelia that are amenable to a variety of electrophysiological, molecular and optical methods for studies of epithelial cell function.
