Abstract

Mast cells are pivotally involved in many diseases with an allergic basis, e.g. asthma and urticaria. In human airways, mast cells constitute 0.5% of the cells lining the bronchi1 where they are ideally situated to interact with inhaled allergens. Evidence for mast cell activation, at least in allergen provocation experiments, derives from the detection of elevated mast cell mediators in bronchoalveolar lavage fluid2 and in blood plasma3,4. Similarly, provocation of urticarial reactions with allergen5 or non-immunological stimuli6–9 is associated with a local elevation of mast cell mediators. The possible therapeutic benefit of drugs which inhibit mast cell mediator release would, therefore, appear to be great. This chapter describes the effects on mast cells of drugs used in the treatment of allergic diseases.
