Abstract
Azospirillum strains examined so far contain large plasmids with which no phenotypic property has been associated. Homology was previously detected between total DNA from several Azospirillum strains and Rhizobium meliloti nodulation (nod and hsn) genes, which in Rhizobium are plasmid-borne. From A. brasilense Sp7, a 10kb EcoRI fragment sharing homology with the R. meliloti hsn region was cloned in pUC18, to yield pAB502. The hsn homologous region was localized on the 90 Mda plasmid, p90, contained in strain Sp7. This was confirmed after mutagenesis of pAB502 by Tn5-Mob and recombination of the insertions into p90. The 10 kb EcoRI fragment was used as a probe to study homology between plasmids from eight A. brasilense strains and from five A. lipoferum strains. Hybridization with a plasmid of about 90 Mda was observed in two A. lipoferum strains and in all of the A. brasilense strains, except in strain R07 where homology was found with a 115 Mda plasmid. The three A. lipoferum strains, which did not hybridize with the probe, did not contain a 90 Mda plasmid.
