Abstract

Cancer can be defined as a disease that is characterised by a failure of the control mechanisms that are involved in cell division. Rapidly proliferating cells have a higher demand for DNA, and consequently require large quantities of purine and pyrimidine nucleotide substrates. Folic acid (1) plays a central role in the de novo biosynthesis of DNA. The coenzyme N5,N10-methylenetetrahydrofolate provides the one carbon unit necessary for the conversion of deoxyuridine monophosphate (dUMP) to thymidylic acid (dTMP).
