Abstract
We designed a screening system for isolation of the plant cDNA clones which restored a growth defect of a yeast ferrous uptake mutant ctr1. We isolated a cDNA clone, temporary designated as SFD1 (Suppressor of Ferrous uptake Defect), from Fe-deficient barley roots which restored the growth defect of ctr1 mutant on iron deplete media containing 1 mM ferrozine and 1 mM mugineic acid. However, this recovering phenotype did not depend on mugineic acid. Sequence analysis revealed that SFD1 cDNA encoded a novel protein whose deduced amino acid sequence had no significant similarity to any known proteins. These results indicated that SFD1 cDNA encoded a novel protein which was likely to be involved in one of the mechanisms of uptake, metabolism and availability of iron or copper in barley roots.
