Abstract

The elucidation of the intracellular mechanism for the processing of double-stranded RNAs was followed by a rapid implementation of RNAi technology as a standard tool in molecular biology and at the same time fostered the research into miRNA biogenesis and function.

This chapter reviews the mechanism of RNA interference and relevant criteria for the application of si/shRNA-mediated gene silencing as a research tool. Then, the role of endogenous miRNA for the regulation of apoptotic cell death in development and disease is illustrated on selected key pathologies. Finally, examples are given for the successful implementation of RNAi for the dissection of apoptotic cell death pathways and in experimental therapeutic approaches using apoptosis mediators as targets.
