An in vitro Assay System for Measuring Both Vascular Permeability and Endothelial Cell Damage

Abstract

An assay system capable of measuring endothelial cell injury was established. The leakage of FITC-labeled bovine serum albumin through endothelial cell barrier was found to reflect the magnitude of endothelial cell injury. In the presence of complement, anti-ox red blood cell anti-serum caused severe injury to cultured endothelial cells derived from bovine carotid artery, followed by enormous increase in the albumin leakage. Exposure of endothelial cell monolayer to 15-HPETE (15-hydroperoxyeicosatetraenoic acid), one of the lipoxygenase metabolites of arachidonic acid, caused dose dependent injury to the endothelial cells. MCI - 186 (3-methyl-l-phenyl-2-pyrazolin-5-one), a radical scavenger, had a remarkable protective effect on such endothelial cell injury.
