Growth and Induction Effects on the Expression of rhTNF-ß

Abstract

The Trp promoter has been widely used in recombinant technology for the over expression of foreign proteins in E. coli [1,2]. The conditions for optimal expression vary considerably among these reports. In this study, the effects of culture conditions, such as medium composition, temperature and concentrations of tryptophan and 3-β-indoleacrylic acid (IAA) on the expression of recombinant human tumour necrosis factor-β (rhTNF-β) in E. coli K12 HB101 pCG402 were investigated. The expression of the protein is under the control of a modified trp promoter, which lacks the attenuator [3].
