Abstract
Glucosaminyl N-deacetylase/N-sulfotransferase-2 (NDST-2) is a modification enzyme involved in heparin/heparan sulfate (HS) biosynthesis. This bifunctional enzyme catalyzes replacement of the acetyl group in GlcNAc with a sulfate group, which is the first modification step following polymerization of the (GlcNAc-GlcA)n precursor of heparin/HS. It is a key step during biosynthesis, as subsequent modifications, including GlcA epimerization and O-sulfation at various positions occur only in the vicinity of N- sulfate groups. Four mammalian NDST isoforms have been identified. At present, the specific roles of the various isoforms involved in heparin/HS biosynthesis are not known. Single orthologs are present in Drosophila melanogaster and Caenorhabditis elogans.
