Blood compatibility of polyacrylonitrile containing ethyleneoxide and dimethylamino side chains

Summary

Polyacrylonitrile copolymer containing ethyleneoxide (EO) and dimethylamino (DA) side chains was synthesized. For the copolymer, the relation between adhesion of platelets on the copolymer surface and the DA content of the copolymer was examined by a “column method,” whereby a platelet suspension was passed through a column of glass beads precoated with the copolymer.

Retention of platelets was 100% irrespective of DA content for the copolymer having almost the same water content. However, when rat serum albumin (RSA) or rat γ-globulin (RγG) was added to the platelet suspension, the retention of platelets significantly decreased and was reduced to 0% when the DA content was low. A similar result was observed when the copolymer surface was previously coated with RSA or RγG, instead of adding RSA or RγG to the platelet suspension.

The adsorption of RSA or RγG to the copolymer surface was examined in a batch system. Adsorption increased remarkably with a decrease in the DA content. It is concluded that the increase in the DA content decreases the adsorption of RSA and RγG on the copolymer surface, resulting in an increase in the adhesion of platelets on the copolymer surface.
