The evolution of MHC class I genes in cattle

Summary

Detailed analysis of cattle MHC class I haplotypes has been carried out using inbred homozygous animals. Transcribed class I genes have been identified and characterised by cDNA cloning, sequence analysis, and transfection/expression studies. The analysis demonstrated that multiple genes are transcribed, and also revealed substantial variation between haplotypes. Phylogenetic analysis of all available BoLA class I cDNA sequences revealed complex evolutionary relationships. While some groupings of sequences may demonstrate a correlation with individual loci, overall it was not possible to define the origin of alleles using this approach. These data indicate that there may be four or more classical class I genes in cattle, with striking differences in the array of these genes expressed in different haplotypes. This situation may reflect the complex origins of modern domestic cattle breeds.

Various approaches are being used to determine the origin and possible functional significance of the observed haplotype variation in cattle. These include mapping of the class I region, assessment of expression levels of different class I gene products, and phylogenetic analysis of class I sequences from related species.
