Die Aufgabe der Aspartylproteasen besteht in der Spaltung von Peptidbindungen. Sie verdanken ihren Namen dem Vorliegen zweier Aspartate, die den Katalyse- Mechanismus bestimmen. Für den Angriff auf die zu spaltende Peptidbindung verwenden sie als Nucleophil ein Wassermolekül, das sie mit den beiden Aspartatresten in geeigneter Weise polarisieren. Gleichzeitig sind diese Reste aber auch für die Stabilisierung des übergangszustands, den Ladungsausgleich und die übertragung von Protonen verantwortlich. Als erstes Mitglied dieser Enzymklasse wurde das Verdauungsenzym Pepsin intensiv untersucht. Es ist im stark sauren Bereich bei pH-Werten zwischen 1 und 5 aktiv. Die erste 3D-Struktur dieser Aspartylprotease wurde bereits Anfang der 1970er-Jahre in der Gruppe von Alexander Fedorov bestimmt. Im humanen Genom erweist sich die Familie der Aspartylproteasen als relativ klein, sie umfasst 15 Mitglieder. Tabelle 24.1 führt einige wichtige Aspartylproteasen auf.
