Die quantitativen Struktur-Wirkungsbeziehungen, QSAR (meist [kju:sar] ausgesprochen, von engl. quantitative structure-activity relationships), versuchen Zusammenhänge zwischen chemischer Struktur und biologischer Wirkung zu erfassen und quantitativ zu beschreiben. Die untersuchten Substanzen sollten aus einer chemisch einheitlichen Serie stammen und müssen am gleichen biologischen Target angreifen. Dort sollten sie einen identischen Wirkmechanismus aufweisen. Mit QSAR-Verfahren kann man z. B. strukturell analoge Hemmstoffe eines bestimmten Enzyms untereinander vergleichen, nicht aber verschiedene Blutdrucksenker, die an unterschiedlichen Zielproteinen mit abweichenden Wirkmechanismen angreifen. Die Korrelation mit den physikochemischen Eigenschaften bezieht sich immer auf relative Wirkstärken in einem Modell, nicht auf unterschiedliche Wirkqualitäten.
