Zusammenfassung
Pluripotente embryonale Stammzellen (ESC) der Maus sind permanente Linien aus der inneren Zellmasse von Blastozysten oder einzelnen Blastomeren von 8-Zell-Embryonen. Sie differenzieren nach Kultivierung in Zellaggregaten (“embryoid bodies”) in Derivate aller 3 Keimblätter, des Endoderms, des Ektoderms und des Mesoderms, so auch in spontan pulsierende Herzmuskelzellen. Diese Herzzellen können sowohl im Zellverband als auch nach enzymatischer Isolierung in Einzelzellen für pharmakologische und elektrophysiologische Untersuchungen eingesetzt werden.
Die differenzierten Herzmuskelzellen sind charakterisiert durch funktionelle Expression von Adreno- und Cholinozeptoren sowie von L-Typ-Ca2+-Kanälen. Die Aktionspotentiale der pulsierenden Herzzell-Aggregate, als auch isolierter Einzelzellen, zeigten die typische Form von embryonalen Herzmuskel- und Sinusknotenzellen.
Elektrophysiologische Untersuchungen ergaben, daß die an isolierten Herzzellen erhobenen Befunde auf Ionenströme mit den chronotropen Effekten an Herzzellen in Aggregaten von “embryoid bodies” miteinander korrelierten.
Im Verlauf der Differenzierung von ESC in Herzmuskelzellen wirkten embryotoxische Substanzen, wie z.B. Retinsäure (RA), während definierter Zeiten (0–2 d, 2–5 d, 5–7 d) der in vitro-Kultur auf die differenzierenden “embryoid bodies” ein.
Retinsäure führte konzentrations- und zeitabhängig zu einer Modulation der Differenzierung in Herz- oder in Skelettmuskelzellen. Die in Anwesenheit von Retinsäure differenzierten Herzmuskelzellen zeigten funktionelle Expression von Adrenozeptoren und Ca2+-Kanälen, jedoch nicht von muskarinen Cholinozeptoren.
Das ESC-Differenzierungssystem wird vorgeschlagen als alternatives Zellmodell für pharmakologische, elektrophysiologische und embryotoxikologische Untersuchungen und damit als Tierersatzsystem für die in vitro-Reproduktionstoxikologie.
