Zusammenfassung

26 schizophrene Patienten mit tardiven Dyskinesien erhielten 10 Tage offen parenteral 200 mg Amantadin-Sulfat. Die Beurteilung erfolgte mittels TDRS. Wegen schwerer Nebenwirkungen und zusätzlicher Verschlechterung des psychopathologischen Zustandsbildes mußten 3 Patienten vorzeitig aus der Studie genommen werden, 1 wegen Unwirksamkeit. Weitere 3 Patienten wiesen leichte bis mäßige Nebenwirkungen auf. Alle 22 Patienten zeigten unter Amantadin-Sulfat eine signifikante Besserung, wobei jene signifikant besser auf die Therapie ansprachen, die weniger als 1 Jahr an TD litten. Frauen sprachen signifikant besser auf die Therapie an als Männer. Die am Stichtag errechneten Chlorpromazinäquivalente hatten keinen signifikanten Einfluß auf den therapeutischen Erfolg. Besserungen zeigten sich vor allem im fazialen Bereich und im Bereich der oberen Extremitäten.

Summary

Treatment of tardive dyskinesia with amantadine-sulphate. In an open study 26 schizophrenic patients with tardive dyskinesia received a parenteral dose of 200 mg of amantadine sulphate for 20 days. Assessment was done by TDRS. Three patients had to be withdrawn from the study because of severe side-effects and deterioration of their mental illness, and one because treatment was ineffective. Another three patients showed mild to moderate side-effects. All 22 patients showed a significant improvement with amantadine sulphate. Those who had suffered from TD for less than 1 year responded significantly better to treatment. Women responded to treatment significantly better than men. The chloropromazine equivalents calculated on the sample day had no significant effect on therapeutic success. The improvements were most noticeable in the face and the upper limbs.
