Zusammenfassung
Dopamin (DA) wurde von Carlsson et al. [1] entdeckt. Der wesentliche und für die nachfolgenden Entdeckungen essentielle Versuch, Mäuse und Kaninchen durch Einwirkung von Reserpin in ihrer Bewegung zu sedieren und diesen Zustand durch Gabe von L-Dopa aufzuheben, wurde von Carlsson et al. [2] durchgeführt. Damit waren die grundlegenden Experimente für die Untersuchungen eines etwaigen Dopamindefizits bei der Parkinson-Krankheit (PK) getan [3]. Die Therapie der PK mit L-Dopa [4] war daher eine Bestätigung der Modulation extrapyramidal-motorischer Effekte durch DA. Vorher hatten schon Sano et al. [5] DA im Gehirn des Menschen regional analysiert, und Degkwitz et al. [6] hatten L-Dopa erstmals bei Schizophrenen angewendet. Unabhängig von der Wiener Gruppe hatten Barbeau, Murphy und Sourkes [7] als Folge verringerter DA-Ausschei- dung im Harn bei PK L-Dopa angewendet und Erfolge speziell beim Tremor beschrieben, während die Wiener Gruppe besonders Akine- sie und Rigor in den Vordergrund des Interesses stellten. Die Verbesserung der therapeutischen Wirkung von L-Dopa durch Einführen unspezifischer Monoaminoxidase (MAO)-Hemmer [8], des Decarboxylasehemmers Benserazid [9], der Dopa-Hochdosierung [10], dopaminerger Agonisten [11, 12], von Amantadin [13] und von selektiven MAO-Hemmern [14] geben die grundsätzlichen Strategien zur Verbesserung der Anti-Parkinson-Therapie wieder.
