Summary
A device for screening of microorganisms is described. It consists mainly of a membrane filter that can be coated on both sides. It is mounted in a holding device in such a way that a layer of agar can be applied to the top side as well as to the bottom side of the filter, wherein one layer of agar is used to receive the effective substance-producing microorganisms, while the other layer of agar contains the test bacilli.
The effective substances, produced by the microorganisms or artificially applied in one layer, diffuse from the one layer of agar into the second layer of agar and become effective there.
The device is especially suitable in basic screening of organisms that produce effective antibiotic substances or that produce amino acids.
Other applications are for example detection of auxotrophs in genetic research, detection of DN-ase-active staphylococci in food industry and medicine, screening of high performance mutants for fermentation in antibiotic and amino acid industry.
Zusammenfassung
Eine Anordnung für das Screening von Mikroorganismen wird beschrieben. Diese besteht in der Hauptsache aus einem Membranfilter, welches von beiden Seiten beschichtet werden kann. Die Membran ist in der Halterung so fixiert, daß eine Agarschicht auf die obere Seite gebracht werden kann und eine zweite Agarschicht auf die untere Seite, wobei die obere Schicht zum Beispiel die wirkstoffproduzierenden Mikroorganismen aufnimmt und die untere Schicht die Testorganismen.
Die Wirkstoffe, die von den Organismen produziert werden oder die auch künstlich aufgebracht werden, diffundieren von einer Agarschicht in die andere und werden dort wirksam.
Diese Anordnung ist besonders für das grundlegende Screening von Mikroorganismen geeignet, die Antibiotika und Aminosäuren produzieren.
Andere Anwendungen bestehen zum Beispiel beim Nachweis von auxotrophen Organismen in der Genetik, beim Nachweis von DN-ase aktiven Staphylokokken in der Nahrungsmittelindustrie und in der Medizin und beim Screening von Hochleistungsmutanten für die Fermentation in der Antibiotika- und Aminosäureindustrie.
