Abstract

The most important changes in the Raman Spectra of nucleic acids, caused by alterations in secondary structure, are reported and discussed. These changes are demonstrated in the case of the polynucleotide polyuridylic acid. It is shown that the effects of base pairing on the carbonyl stretching vibrations of the nucleic acid bases can be observed in H2O solution by subtracting the Raman scattering of the solvent. The advantages of Raman spectroscopy in the study of nucleic acid conformations are demonstrated in the case of the double helical complex polyadenylic acid × polyuridylic acid, of various transfer ribonucleic acids and of the 3′,5′-ribodinucleoside monophosphates.
