Zusammenfassung

Der Anlaß für unseren Arbeitskreis, sich mit dem Problem der Kollagenstruktur zu befassen, war ein äußerer. Die Kollagenfaser ist das hauptsächliche Bauelement des Bindegewebes und damit auch der Haut und des Leders. Zunächst haben wir uns — und dies gilt schon von meinem Vorgänger Max Bergmann — das Problem der chemischen Struktur des Kollagens viel zu einfach vorgestellt: Die Molekülkette, von der es zunächst offen schien, ob sie in der Länge begrenzt sei oder „fakultativ endlos“ wie die der Cellulose, sollte aufgebaut sein aus Sequenzen von je 3 bzw. 6 Aminosäuren, unter denen Glycin, Prolin und Hydroxyprolin regelmäßig wiederkehren sollten (27, 12). Im Gegensatz zu den rein spekulativen Vorstellungen über die Aminosäureanordnungen anderer Proteine, die damals auf ähnlicher Grundlage entwickelt worden sind, hat sich im Falle des Kollagens die Bergmannsche Vorstellung einer periodischen Wiederkehr kurzer, gleichartiger Sequenzen als fruchtbar erwiesen. Sie war wegleitend für die späteren 3-Ketten-Helix-Modelle nach Pauling (316), Ramachandran (334, 335, 337) und Rich und Crick (345), und sie hat in den chemischen Untersuchungen, über die unten berichtet wird, zum mindesten für die „apolaren“ Bereiche eine Bestätigung gefunden.
