Zusammenfassung
Umfangreiche Untersuchungen der letzten Jahre haben gezeigt, daß an der Generierung epileptischer Aktivität ein pathologischer exzessiver Calciumionenstrom entscheidend beteiligt ist (vgl. Speckmann et al. 1986). Dieser Calciumionenstrom ist durch Substanzen, die Calciumkanäle blockieren können, zu unterdrücken, so daß in der Folge auch epileptische Aktivität durch diese Calciumkanalblocker vermindert werden kann (cf. Walden et al. 1992). Darüberhinaus konnte gezeigt werden, daß das Antiepileptikum Carbamazepin ebenfalls calciumantagonistische Eigenschaften aufweist (Walden et al. 1992, 1993). Diese Substanz wird in der Psychiatrie seit vielen Jahren zur Behandlung des manischen Syndroms und zur Prophylaxe affektiver Störungen eingesetzt. An der Wirkung dieser Substanz bei den genannten psychischen Erkrankungen kann der calciumantagonistische Effekt von großer Bedeutung sein, da in der Pathophysiologie affektiver Psychosen eine gestörte intrazelluläre Calciumionenhomöostase diskutiert wird (vgl. Dubovsky 1993). Dementsprechend ergibt sich die Frage, ob reine Calciumantagonisten auch in der Behandlung affektiver Störungen eingesetzt werden können.
