Kürzlich wurde von uns ein neuer Pyrogentest vorgeschlagen (Hartung T. und Wendel A., 1995), der darauf beruht, daß Zusatz von fiebererzeugenden Substanzen (Pyrogenen) zu menschlichem Blut hochsensitiv zur Freisetzung derjenigen Botenstoffe führt, die im Organismus die Fieberreaktion auslösen. Damit macht sich das Verfahren die primäre Fieberreaktion des Menschen zunutze. Die für die Fieberreaktion verantwortlichen Immunzellen liegen in ihrer natürlichen Umgebung vor, wodurch Präparationsartefakte weitgehend ausgeschlossen sind.

Die Testentwicklung ist im weitesten abgeschlossen. Das Paul-Ehrlich-Institut, D-Langen, plant, im Rahmen der Evaluierung und Prävalidierung, diesen Test zur Chargenprüfung von Blutprodukten zu übernehmen und parallel zu dem im Hause durchgeführten Kaninchen-Pyrogentest einzusetzen. Daran kann sich ein Ringversuch zur Validierung der Methode anschließen, mit dem Ziel der Anerkennung als Ersatzmethode für den Kaninchen-Pyrogentest.

Summary

A pyrogen assay based on human whole blood to replace the rabbit pyrogen test

Recently, a pyrogen detection system was introduced that takes advantage of the fact that in human whole blood, minute amounts of pyrogens initiate the formation of mediators causing fever in the organism. Thus, this in vitro method makes use of the primary fever inducing reaction of humans. In contrast to the Limulus assay, it is not restricted to the detection of endotoxins derived from Gram-negative bacteria. All blood cells, including leukocytes, are kept in their natural environment and thus major preparation artefacts are minimized. The authority in charge of safety of parenteralia in Germany, the Paul-Ehrlich-Institute, D-Langen, plans to evaluate and prevalidate this assay in comparison to the standard rabbit pyrogenicity test. A validation study will follow with the aim to replace the animal test.
