Mouse ovarian follicle culture: Can this be a useful system for evaluating potential ovarian toxic substances?

Summary

A major advantage of the intact follicle unit as test system is that all competent cell types which support the developement of the oocyte are present: In a simplified and controlled environment, this allows that treatments which influence in vivo fertility can be evaluated in vitro while maintaining a strong relationship to in vivo events. Although at a very early stage, results of follicle culture in the mouse to evaluate the effects of ovatian toxins suggest that follicle culture will make possible the study of mechanism of action on the isolated follicle unit over a range of developmental stages.

Zusammenfassung

Mausfollikelkulturen: ein Evaluierungssystem für potentiell toxische Substanzen?

Der intakte Follikel ist eine physiologische Einheit, die alle kompetenten Zellarten enthalt, die fair die Follikelreifung wichtig sind. Wenn Follikelkulturen zu Testzwecken in der Toxikologie eingesetzt werden, haben sie den Vorteil, daβ sich die Kulturbedingungen leicht standardisieren lassen and zwar unter Bedingungen, die denen in vivo sehr ähnlich sind. Deshalb ist es mit dem Testsystem möglich, den Einfluβ von toxischen Stoffen auf die Befruchtung unter Verhaltnissen zu untersuchen, die denen in vivo weitgehend Mnlich sind. Obwohl das Testsystem erst seit kurzer Zeit eingesetzt wird, um Umweltchemikalien mit östrogenen Eigenschaften zu erfassen, zeigt sich bereits jetzt, daβ Follikelkulturen ein wertvolles in vitro-Kultursystem in der Reproduktionstoxikologie sind, das noch weiterentwickelt and validiert werden muβ.
