Zusammenfassung
Das Hepatitis-B-Virus ist ein doppelsträngiges DNA-Virus, das zur Familie der Hepadnaviridae gehört. Es ist das kleinste bekannte DNA-Virus mit nur 3200 Basenpaaren im Genom. Dieses besteht aus einem Minusstrang, der für die strukturellen Proteine Pre-S, S und Core sowie die replikativen Proteine Polymerase und X-Protein kodiert und fast zirkulär geschlossen ist, und einem Plusstrang, der kürzer ist. Im Inneren des Virus befindet sich das Nucleocapsid bestehend aus Genom, HBcAg, Polymerase. Die Oberfläche enthält Untereinheiten, die aus dem S-Protein, dem S- und PreS1-Protein oder PreS1-, PreS2- und S-Protein bestehen. Pre-S Proteine spielen wahrscheinlich eine Rolle bei der Bindung des Virus an die Leberzelle. Im Blut zirkulierende 22-nm-Partikel sind sphärische oder tubuläre HBsAntigenpartikel. Infektiöse Dane-Partikel messen 43 nm. Die a-Determinante des HBs-Antigen ist das neutralisierende Epitop, das gegen alle Subtypen einen Schutz verleiht. Andere Determinanten besitzen diese Eigenschaft nicht (d, y und w, r) und bilden in wechselnder Kombination die Subtypen adw, adr, ayw, ayr.
