Summary
Primary and permanent fish cells do not only represent an excellent alternative to conventional fish tests in aquatic (eco)toxicology, but also provide a tool for studies on the mechanisms of toxicity of chemical substances. Whereas permanent fish cell lines are particularly suitable for routine toxicity testing, primary hepatocyte cultures have proved particularly useful for morein- depth studies in the underlying mechanisms of toxicity. The present communication gives examples for the potential use of toxicological studies with fish cell culture systems with particular emphasis on (1) routine toxicity testing of sewage water and related environmental samples, (2) the use of fish cell culture systems in genotoxicity testing, (3) the use of fish cells in biotransformation studies, (4) the combination of cytotoxicity testing with bioassay-directed chemical fractionation (TIE), and (5) the toxicological analysis of sediments and suspended matter in aquatic ecosystems. On the basis of the data provided, a multi-tiered test schedule for the evaluation of the biological activity of xenobiotics and potentially contaminated field samples will be presented, which should allow for an at least 90% reduction of the number of fish used in (eco-)toxicological routine toxicity testing.
Zusammenfassung
Primäre Fischzellen und permanente Fischzellinien stellen nicht nur ein ausgezeichnete Alternative zu konventionellen Fischtests in der aquatischen (Öko-)Toxikologie dar, sondern bieten auch die Möglichkeit, Mechanismen der Toxizität von chemischen Verbindungen zu untersuchen. Während sich permanente Fischzellinien vor allem für Routinetoxizitätstests eignen, haben sich Primärzellkulturen von Hepatocyten für weiterführende Untersuchungen zu den Mechanismen, die einer toxischen Wirkung zugrunde liegen, als geeignet erwiesen. Die vorliegende Arbeit gibt eine Zusammenstellung möglicher Einsatzgebiete für toxikologische Untersuchungen mit Fischzellkulturen, wobei das Schwergewicht auf folgende Gebiete gelegt wird: (1) die Bestrebungen, Fischzellkultursysteme als Ersatzmethode in Routinetoxizitätsprüfungen von Abwässern und anderen Proben aus der Umwelt einzuführen, (2) den Einsatz von Fischzellkulturen in Genotoxizitätsprüfungen, (3) die Problematik der Biotransformation in toxikologischen Tests mit Fischzellkulturen, (4) den Einsatz von Toxizitätstests mit Fischzellkulturen in integrierten Teststrategien wie der Bioassay-dirigierten Fraktionierung und (5) Möglichkeiten des Einsatzes von Fischzellkulturen zur toxikologischen Bewertung von Sedimenten. Auf der Basis der vorgestellten Daten wird schließlich ein mehrstufiges Testsystem für die Bewertung der biologischen Aktivität von Xenobiotika und kontaminierten Umweltproben entwickelt, das eine Reduktion der in Routinetoxizitätsprüfungen eingesetzten Fische um mindestens 90% gestatten sollte.
