Zusammenfassung
Wesentliche Voraussetzung zum Verständnis des Wirkmechanismus eines Pharmakotherapeutikums ist die Kenntnis über Aufnahme und Verteilung im Organismus. Als Zwischenstufe zwischen in vitro-Untersuchungen an Tumorzellen und den Untersuchungen im Tierversuch etablierten wir die Chorioallantoismembran des bebrüteten Hühnereies (CAM) als in vivo-Tumormodellsystem. Dieses Modell bietet den Vorteil, vaskularisierte Mikrotumore in Echtzeit beobachten zu können. Untersucht wurde die Aufnahme und Verteilung verschiedener fluoreszierender Tumortherapeutika in Blasenkarzinomen auf der CAM mit Hilfe konfokaler Laserscanning Mikroskopie. Die Applikation der Substanzen erfolgte sowohl topisch als auch intravasal. Die daraus gewonnenen Ergebnisse konnten mit Ergebnissen aus in vitro- Untersuchungen an Tumorzellen korreliert werden. Darüberhinaus konnte aufgrund der im CAM-Modell gewonnenen Daten bereits vorliegende Untersuchungsergebnisse in Rattenversuchen besser interpretiert und weitere notwendige Tierversuche präzise formuliert werden. Aufgrund dieser Ergebnisse konnte gezeigt werden, daß sich das CAM-Modellsystem hervorragend als Screening-Verfahren eignet, um Tierversuche exakt zu planen. Dadurch kann die Anzahl der Versuchstiere stark eingeschränkt werden.
Summary
Prerequisite for understanding pharmacodynamics is the knowledge about uptake and biodistribution in an organism. As an intermediate stage between in vitro investigations in tumor cells and animal experiments, we established the chick chorioallantoic membrane of the fertilized hen’s egg (CAM) as an in vivo model system. This model system has the advantage of real time Observation of vascularized microtumors. We studied uptake and distribution of different fluorescent anticancer drugs in bladder tumors on the CAM by confocal laser scanning microscopy. Drugs were applicated topically as well as intravenously. The results of these experiments could be correlated with results received from in vitro tests in tumor cells. In addition the data found in the CAM model could help to interpret results we found in rat experiments and to plan precisely further necessary animal tests. Based on these results the chorioallantoic membrane system proved to be an excellent Screening system for exactly planning of animal tests and by it for reduction of the number of test animals.
