Zusammenfassung
Tacrin (1,2,3,4-Tetrahydro-9-AcridinaminMonohydrochlorid, Abb. 12.1.1) ist ein zentral wirksamer noncompetitiver reversibler Cholinesterasehemmer. Der durch Tacrin bedingte Anstieg von Acetylcholin soll den im Rahmen der Demenz vom Alzheimer Typ (AD) auftretenden Acetylcholinmangel (Acetylcholinmangel-Hypothese der Demenz) kompensieren. Man geht davon aus, daß Tacrin neben der katalytisch wirksamen Stelle an das Accetylcholinesterase-Molekül bindet und dadurch die Enzymaktivität inhibiert (Davis and Powchnik 1995). In hohen Konzentrationen weist Tacrin andere Effekte auf, wie eine Blockade von Natrium- und Kaliumkanälen (Freeman and Dawson 1991), Veränderungen im monoaminergen Metabolismus (Adem et al. 1989) und von monoaminerger Aufnahme (Druckarch et al. 1988).
