Zusammenfassung
Das CFTR-Gen, das für die Synthese des Cystic Fibrosis Transmembrane Conductance Regulator Proteins verantwortlich ist, wurde 1989 erstmalig kloniert und sequenziert (Riordan et al. 1989; Kerem et al. 1989), nachdem es schon vorher auf dem langen Arm des Chromosoms 7 imAbschnitt q31-32 lokalisiert werden konnte. Es handelt sich um ein sehr großes Gen mit einer Länge von ca. 230 kB. Das RNA-Transcript hat eine Länge von 6,5 kB und codiert für ein Protein von 1480 Ami-nosäuren. Das CFTR-Gen besteht aus 27 Exons mit 26, z.T. sehr großen dazwischenliegenden Introns. Es gehört zur Genfamilie der ABC-Transporterproteine (ABC = ATP-Binding-Casette). Das CFTR-Protein, ein cAMP-regulierter Chloridkanal, zeigt den für diese Proteinfamilie typischen Aufbau aus 2 Transmembrandomänen bestehend aus je 6 membrandurchspannenden Segmenten, 2 nucleotidbindenden und einer großen, für das CFTR-Gen spezifischen, regulatorischen Domäne. Der Zusammenhang zwischen den Exons und den Proteindomänen ist in Abb.1 Darge-stellt.
