Cytochemistry of the Nucleic Acids

Abstract

Cytochemical methods with quantitative aims are now standard procedures in many laboratories. Following a period of technical progress, quantitative cytochemistry has entered a phase in which the investigation of a cell biological problem, sometimes with direct medical import, is the main goal. A wider use of the methods is welcome, but the need for careful attention to sources of error and limits of accuracy, cannot be too firmly stressed. By far the most encouraging sign is the small but increasing use in combination of two or more methods which give similar information. In particular, a combination of microspectrophotometry, or biochemical methods, with autoradiographic detection of isotopically labelled precursors, has the advantage of reciprocal checking in addition to providing complementary data. Fortunately, the undisputed biological importance of desoxyribose nuclei acid (DNA) is matched to some extent by the number and efficiency of the methods available for its assay. If it had turned out that lipids, for example, were the basis of the genetic material, the lack of quantitative methods would have hampered progress. While it may be argued that methods would be developed, it is significant that we are still without a satisfactory quantitative cytochemical method for the estimation of RNA. Quantitative methods for DNA were available before its biological significance was generally accepted.
