Cytokine Genes as Molecular Adjuvants for DNA Vaccines

Abstract

DNA vaccines encoding a foreign antigen induce antigen-specific cellular and humoral immune responses to protect against viral and bacterial infections, parasites, cancers, and autoimmune diseases. However the relatively low efficacy of DNA vaccinations in large animal species and humans has hindered their practical use as therapeutics. Therefore, there is an urgent need to improve protective responses generated by DNA vaccines. Among various improvement strategies, the incorporation of cytokine expressing plasmids as molecular adjuvants have been widely studied in the past years, yet still without significant clinical application. Cytokines play critical roles in natural immune responses during the presentation of antigens, and in particular, in directing T-cell polarization. This chapter reviews recent progress in the co-application of cytokine-encoding genes used for enhancement and direction of immunogenicity, as well as discusses their therapeutic potential for future applications.
