Abstract
In the present study the effect of lipid-loaden macrophages on SMC proliferation was investigated in an indirect co-culture system. The cells, SMC and macrophages, were separated by a membrane which allows diffusion of molecules but no cell-cell-contact. Mouse peritoneal macrophages (MPM) and the macrophage-like cell-line U937 were incubated with oxidized and acetylated LDL and co-cultivated with rat SMC.
It could be demonstrated that macrophages loaded with modified LDL and transformed to foam cells have a direct effect on SMC by increasing cell proliferation.
Zusammenfassung
Der Einsatz von Zellkulturmodellen zur Erforschung pathologischer Vorgänge und möglicher pharmakologischer Therapieansätze gewinnt mehr und mehr an Bedeutung. Zur Gewährleistung der Übertragbarkeit auf Organe bzw. den Gesamtorganismus ist eine hohe Komplexität der in-vitro-Testsysteme notwendig, um Einblick in die vielfältigen Stoffwechselprozesse und die damit verbundene interzelluläre Kommunikation zu erhalten. Dazu bedarf es einer Erweiterung der bisher üblichen eindimensionalen Modelle und Monokultursysteme.
