Abstract

In spite of their widespread use there was limited information on the pharmacokinetics of diuretics until recently. This was due to the absence of suitable methods for the quantification of these drugs. There was a lack of sensitivity and specifity in measuring the intrinsic fluorescence in the cuvette (furosemide) and in the derivatisation to coloured products (hydrochlorothiazide, furosemide). During the last few years methods have been developed using GLC and HPLC to separate the drug from plasma constituents. Quantitative thin-layer chromatography has also proved to be successful for measurement of drugs in biological material.
