Zusammenfassung
Die meisten Proteine sind in Zellen, Geweben und anderen biologischen Materialien lokalisiert. Werden Proteine aus dieser natürlichen Umgebung entfernt, können sie sehr schnell ihre biologische Aktivität verlieren. Sie müssen zur weiteren Aufarbeitung in Lösung gebracht (solubilisiert) werden. Dazu dienen meist Pufferlösungen mit Ionenstärken und pH-Werten, bei denen die Proteine stabil sind.
