Zusammenfassung
Die Rolle von Apolipoproteinen (Apo A IV) im Lipidstoffwechsel ist noch nicht völlig geklärt. Aus der Literatur gibt es zunehmend Hinweise dafür, daß es eine Rolle im Cholesterinrücktransport spielt. Um dies näher zu untersuchen, haben wir seine Fähigkeit untersucht, aus cholesterinbeladenen Mausadipozyten zellulären Cholesterinausstrom zu vermitteln. Wenn diese cholesterinbeladenen Zellen mitApoA-IV-haltigen Phospholipidkomplexen bei 37 Grad inkubiert wurden, kam es zu einem zeit- und konzentrationsabhängigen Cholesterinausstrom aus den Zellen. Die Konzentration von Apo A IV/Phospholipidkomplexen zur Vermittlung halbmaximalen Ausstroms betrug 0,3 × 10-6 M. Bindungsexperimente, durchgeführt an intakten Zellen bei 4° C mit Phospholipidkomplexen, die radioaktiv markiertes Apo A IV enthielten, zeigten, daß spezifische A-IV-Bindungsstellen existierten mit einem Kd von 0,32 × 10-6 M. Kompetitive Bindungsexperimente mit Phospholipidkomplexen, die entweder radioaktiv markiertes A IV, A I oder A II enthielten, sprachen dafür, daß alle drei Apolipoproteine von den gleichen Zelloberflächenstrukturen erkannt wurden. Wir schließen daraus, daß neben Apo A I auch Apo A IV, das in der interstitiellen Flüssigkeit in ähnlichen Konzentrationen vorkommt — wie hier in vitro eingesetzt — auch in vivo eine bedeutende Rolle im Cholesterinausstrom aus peripheren Zellen spielt.
