Abstract
At last year’s conference of the German Society of Arteriosclerosis in Blaubeuren, our team presented an animal model, which allows by intravital fluorescence microscopy the demonstration of leukocyte adhesion to the endothelium of arterioles and venules in response to intravenous injection of oxidized human low density lipoproteins (oxLDL). In order to further validate the model and to provide further evidence for the pathogenetic role of oxLDL in atherogenesis, the following questions were addressed:

Are superoxide radicals involved in oxLDL-induced leukocyte adhesion? This question was answered in experiments, in which pretreatment of hamsters with a low dose of CuZn-superoxide dismutase (SOD, 0.25 mg/kg) or heparin (2000 U/kg; each drug administered 10 minutes prior to oxLDLinjection) significantly attenuated oxLDL-induced leukocyte adhesion.

Is oxLDL-induced leukocyte adhesion confined to the microcirculation? Using scanning electron microscopy, we have now demonstrated leukocyte adhesion to the endothelial lining of aortas of hamsters 15 minutes after injection of oxLDL.

Are CD11b/CD18 adhesion receptors involved in oxLDL-induced leukocyte adhesion?
Answers to this question were obtained from flow cytometric studies, demonstrating up-regulation of CD11 b on human PMN and monocytes 30 minutes after in vitro stimulation with oxLDL, and from experiments on nude mice, demonstrating a complete inhibition of oxLDL-induced leukocyte adhesion to the microvascular endothelium by pretreatment with functionally blocking monoclonal antibodies against the CD11 b subunit of CD11 b/CD18.
Zusammenfassung
Auf der Jahrestagung der Deutschen Gesellschaft für Artherioskleroseforschung in Blaubeuren im letzten Jahr wurde von unserer Arbeitsgruppe ein Tiermodell vorgestellt, in dem intravitalmikroskopisch die Adhäsion fluoreszenzmarkierter Leukozyten am mikrovaskulären Endothel eines Hautmuskels als Reaktion auf die intravenöse Injektion oxidativ modifizierter humaner Low Density Lipoproteine (oxLDL) visualisiert werden kann. Um das Modell bzw. die pathogenetische Rolle von oxLDL im Rahmen der Atherogenese weiter zu charakterisieren, wurden drei weitere Fragestellungen experimentell untersucht.

An der durch oxLDL stimulierten Leukozytenadhäsionssteigerung sind Superoxidradikale beteiligt.
Dieser Schluß ergibt sich aus Versuchen, in denen eine Vorbehandlung der Hamster mit einer niedrigen Dosis Superoxiddismutase (CuZn-SOD, 0,25 mg/ kg KG, i.v.) bzw. Heparin (2 000 IE/kg KG, i.v., welches endothelial gebundene extrazelluläre Superoxiddismutase in die Zirkulation freisetzt) zu einer signifikanten Reduktion der durch oxLDL stimulierten Leukozyten/EndothelInteraktion führte.

Die Injektion von oxLDL führt zur Leukozytenadhäsionssteigerung nicht nur am mikrovaskulären Endothel, sondern auch am Endothel großer Arterien. Dieses Phänomen konnte mittels Rasterelektronenmikroskopie des Aortenendothels von Hamstern nach Injektion von oxLDL nachgewiesen werden.

OxLDL führt zu einer Expressionssteigerung von Adhäsionsrezeptoren CD11 b/CD18 auf Granulozyten und Monozyten.
Dieses Ergebnis beruht auf der durchflußzytometrisch quantifizierten Rezeptorexpression nach Inkubation von humanem Vollblut mit heterologem oxLDL. Die Vorbehandlung des Vollblutes mit dem membranfluidisierenden Agens N-Butanol verhinderte die oxLDL-induzierte Rezeptorexpressionssteigerung, was auf eine Regulation der Expression von Adhäsionsrezeptoren auf Leukozyten durch die Plasmamembranfluidität hindeutet.
