Abstract
Fibromuscular plaques were produced with daily repeated electrical stimulation over a period of 28 days in the carotid artery of rabbits. After a pause of 28 days without any influence of atherogenetic stimuli nicotine containing osmotic minipumps were implanted (3 µg/kg/min). After a period of either 7 days or 14 days the mitotic activity of smooth muscle cells and endothelial cells of the plaques were determined using bromo-deoxy-uridine and antibodies specific for smooth muscle cells or endothelial cells.
Zusammenfassung
Bereits 1911 wurde unter Claudicatio intermittens-Patienten eine drei- bis fünffache Häufung von Rauchern beobachtet. In der Zwischenzeit gibt es keinen Zweifel mehr daran, daß Rauchen einer der Risikofaktoren der Arteriosklerose ist [6, 7, 10]. Ein wichtiger Bestandteil des Zigarettenrauches ist das Nikotin. Seine potentiell schädigende Wirkung auf Endothel- und glatte Muskelzellen wurde nachgewiesen [3, 7, 10]. In dieser Studie wurde untersucht, ob eine direkte Wirkung von Nikotin auf das Proliferationsverhalten glatter Muskelzellen und Endothelzellen in vivo nachzuweisen ist und ob präformierte Gefäßwandplaques durch Nikotineinwirkung in ihrem weiteren Wachstum stimulierbar sind.
