Abstract
Not only the chylomicrons, but also the VLDL triglycerides increase following oral fat intake. Size fractionation of the plasma lipoproteins, allowing a separation of the chylomicrons, chylomicron remnants and VLDL, together with determination of retinyl palmitate in these fractions, shows that a large part of the VLDL does not contain retinyl palmitate of oral origin. This VLDL must therefore derive from the liver. One possibility to explain a postprandial increase in the synthesis of VLDL could be an increased input of free fatty acids. The postprandial heparin-induced release of large quantities of free fatty acids showed a very heterogeneous time course in 8 test persons. However, there was a close correlation between the amplitude and duration of free fatty acids on the one hand, and the steady reincrease of the VLDL-triglycerides on the other. Since the hepatic lipoproteins which are rich in triglycerides persist considerably longer than the chylomicrons, this finding is significant in regard to the potential atherogenicity of postprandial lipoproteins. Elevated levels of these lipoproteins induce a lipid transfer which leads to a reduction of the HDL-cholesterol and at the same time to an increase in the cholesterol content of the VLDL. Both circumstances are considered to be atherogenic.
Zusammenfassung
Nach oralem Fettverzehr steigen nicht nur die Chylomikronen-, sondern auch die Very low density lipoprotein(VLDL)-Triglyzeride an. Eine Größenauftrennung der Plasmalipoproteine, welche die Trennung von Chylomikronen, ChylomikronenRemnants und VLDL erlaubt, zeigte in Kombination mit der Retinylpalmitatmessung in den Fraktionen, daß ein Großteil der VLDL kein oral zugeführtes Retinylpalmitat enthält und somit aus der Leber stammen muß. Als eine Möglichkeit der postprandialen Synthesesteigerung von VLDL kommt ein verstärkter Zufluß freier Fettsäuren in Frage. Die postprandiale heparininduzierte Freisetzung großer Mengen freier Fettsäuren zeigte bei acht Versuchspersonen sehr heterogene Verläufe, aber eine enge Korrelation von Höhe und Dauer der Spiegel der freien Fettsäuren einerseits und anhaltendem Wiederanstieg der VLDL-Triglyzeride andererseits. Da hepatische triglyzeridreiche Lipoproteine wesentlich länger persistieren als Chylomikronen, ist dieser Befund für die potentielle Atherogenität postprandialer Lipoproteine bedeutsam. Hohe Spiegel dieser Lipoproteine induzieren einen Lipidtransfer, der zu einer Verminderung des High density lipoprotein(HDL)-Cholesterins und gleichzeitig zu einer Anreicherung der VLDL mit Cholesterin führt. Beide Umstände gelten als atherogen.
